Shifted Termination Assay (STA)

Platform Description
Shifted Termination Assay (STA) is a proprietary technology that uses uniquely designed primers, a mixture of modified enzymes and specially synthesized nucleotides. The STA reaction recognizes wild type or mutant target sequences and selectively extends detection primers with 1 to 20 labeled nucleotides. This extension is repeated 20 times to enrich the mutation signal. The enriched mutation signals are then further amplified by an enzymatic color reaction and will result in a strong color.

STA reaction is repeated 20 times to enrich mutation signal. The mutation signal is then further amplified by enzymatic reaction in colorimetric detection. Studies using a combination of wild type and mutant DNA show that the enrichment process enhances sensitivity, enabling the assay to detect mutations as low as 1%.
